Tilda Swinton already played Orlando! It was in a film all about Orlando, called Orlando. 
Someone suggested Cate Blanchett once, but personally, I'd choose British model-turned-actress Lily Cole for the female Orlando.
And as for the male Orlando... who could play him? Perhaps if Hugh Jackman shaves off that beard...
